The combination of pertuzumab and trastuzumab is used to treat HER2-positive breast cancer, an aggressive form of cancer that overexpresses the human epidermal growth factor receptor 2 (HER2). This targeted therapy is indicated for both advanced and early stages of the disease, in combination with chemotherapy.
Pertuzumab and trastuzumab are monoclonal antibodies that work synergistically to block HER2 receptor signaling. Pertuzumab inhibits the dimerization of HER2 with other HER receptors, while trastuzumab blocks HER2 receptor activation and intracellular signaling. This dual blockade reduces tumor cell growth and proliferation.
This combination is used in neoadjuvant treatment (before surgery) to reduce tumor size, in adjuvant treatment (after surgery) to prevent recurrence, and in metastatic cancer treatment to prolong survival.
Side effects may include diarrhea, nausea, fatigue, hair loss, decreased appetite, and infusion-related reactions. In rare cases, more severe effects such as heart failure or severe allergic reactions may occur. Regular monitoring of cardiac function is essential during treatment.
