About this active substance

Quizartinib is a targeted cancer medicine used in adults with newly diagnosed acute myeloid leukaemia with the FLT3-ITD mutation. This mutation can drive cancer cell growth. Quizartinib blocks FLT3 signalling and is given with standard chemotherapy, then may continue alone as maintenance treatment according to the haematology plan.

It is taken by mouth, usually once daily during specific parts of chemotherapy cycles and later according to the maintenance schedule. The dose may be adjusted based on blood tests, electrocardiograms and tolerability. Do not crush the tablets and do not change the dose yourself. If a dose is missed, follow the instructions from your doctor or the leaflet.

Side effects may include nausea, vomiting, diarrhoea, abdominal pain, tiredness, reduced appetite, infections, fever and changes in blood counts. A key safety issue is prolongation of the QT interval on the ECG, which can increase the risk of abnormal heart rhythms. Seek medical help if you have fainting, palpitations, severe dizziness, fever, bleeding or signs of infection.

Before and during treatment, ECGs and checks of potassium and magnesium are needed. Tell your doctor about all medicines you use, including antifungals, antibiotics and anti-sickness medicines, because some can increase heart risk. Quizartinib may harm an unborn baby, so effective contraception is required; discuss breastfeeding. Treatment must be managed by an experienced leukaemia team.
